ABSTRACT

To investigate the N-containing compounds in the seeds of Paganum harmala,fourteen compounds were finally isolated from the 95% Et OH extract of P. harmala seeds by using various chromatographic techniques including silica gel,MCI resin,and ODS column chromatography as well as the semi-preparative HPLC. Depending on spectroscopic techniques and comparison with the reported data in the literatures,the structures of all compounds were identified as N-[3-(2-amino-4-methoxyphenyl)-3-oxopropyl]acetamide(1),dehydroharmalacidine(2),harmalacidine(3),harmine N-oxide(4),harmine(5),tetrahydroharmine(6),demethylharmalacidine(7),harmol(8),tetrahydroharmol(9),harmindol β-D-glucopyranoside(10),tryptophyl β-D-glucopyranoside(11),pegamineβ-D-glucopyranoside(12),vasicol(13) and vasicinone(14). Among them,1 was a new compound,and 2 and 10 were obtained as natural products for the first time.

ABSTRACT

A total of twenty-two compounds were isolated from the 95% EtOH extract of Eclipta prostrata by various purification steps, and their structures were established as ecliptalignin A (1),ecliptasaponin Ⅰ (2), ecliptasaponin Ⅱ (3), echinocystic acid (4), 3-oxo-16α-hydroxy-olean-12-en-28-oic acid (5), acacetin-7--rutinoside (6), luteoloside (7), apigenin (8), luteolin (9), acacetin (10), skullcapflavone Ⅱ (11), kaempferol (12), kaempferide (13), quercetin (14), 4',7-dihydroxyl-3',6'-dimethoxylisoflavone-7--glucoside (15), ecliptal (16), 5-hydroxymethyl-(2,2',5',2″)-terthienyl tiglate (17), psoralen (18), isopsoralen (19), wedelolactone (20), crinumaquine (21), and 2,3,9,12-tetramethoxyprotoberberine (22) mainly based on the spectroscopic techniques, of which 1 was a new lignin analogue, and 5, 6, 10-13, 15, 18, 19, 21 and 22 were isolated form this plant for the first time.

ABSTRACT

Objective :To explore influence of dexmedetomidine (DEX) on coronary artery diameter and myocardial protection in patients with AMI undergoing PCI .Methods :A total of 92 AMI patients ,who received PCI in our hospital from Mar 2016 to Mar 2017 ,were selected .Patients were randomly and equally divided into routine PCI group and DEX group (received DEX intravenous pump based on routine PCI group ).Coronary artery diameter , HR ,SBP ,DBP ,serum levels of cardiac troponin I (cTnI) ,heart type fatty acid-binding protein (H-FABP) and creatine kinase isoenzyme MB (CK-MB) before ,6h and 24h after treatment ,and incidence of adverse reactions were observed and compared between two groups .Results :Compared with routine PCI group ,on 6h and 24h after treatment ,there were significant reductions in coronary artery diameter [24h : (2.68 ± 0.36) mm vs .(2.44 ± 0.35) mm] , HR [24h :(86.56 ± 8.12) beats/min vs .(75.42 ± 7.05) beats/min] ,DBP [24h :(76.26 ± 10.30) mmHgvs.(68.28 ± 9.25) mmHg] and SBP [24h :(133.17 ± 23.76) mmHg vs.(118.28 ± 10.26) mmHg] in DEX group , P<0. 01 all.Compared with before treatment group ,there were significant rise respectively in serum levels of cTnI and CK-MB in two groups on 6h and 24h after treatment ,and serum H-FABP levels significantly decreased respectively , P=0.001 all.Compared with routine PCI group ,on 6h and 24h after treatment ,there were signifi-cant rise in serum levels of cTnI [24h :(1.23 ± 0.05) ng/mlvs.(1.64 ± 0.07) ng/ml] and CK-MB [24h : (125.09 ± 13.16) μg/ml vs.(149.71 ± 14.53) μg/ml] ,and significant reduction in serum H-FABP level [24h : (10.44 ± 2.59) ng/ml vs.(8.31 ± 2.16) ng/ml] in DEX group , P=0.001 all.Incidence rate of adverse reactions in DEX group was significantly lower than that of routine PCI group (6.52% vs.23.91%) , P=0. 020. Conclusion : Though dexmedetomidine can constrict coronary artery ,but it can perform myocardial protection through reducing blood pressure and HR etc .in AMI patients undergoing PCI ,and its adverse reactions are few ,which is worth extending .

ABSTRACT

Objective: To study the chemical constituents in the dried roots of Panax notoginseng. Methods: The constituents were isolated and purified with chromatographic methods. Their structures were elucidated by spectroscopic methods (1D, 2D NMR, UV, IR, [α]D, and HRESI-TOF-MS) and chemical analyses. Results: Twenty saponins including 20(S)-ginsenoside Rh1 (1), 6-O-β-D-(6'-acetyl)-glucopyranosyl-24-ene-dammar-3β, 6α,12β,20S-tetraol (2), ginseno- side Rf (3), notoginsenoside R2 (4), ginsenoside Rg2 (5), ginsenoside Rg1 (6), notoginsenoside Rt (7), koryoginsenoside R1 (8), 6-O-(β-D-glucopyranosyl)-20-O-(β-D-xylopyranosyl)-3β,6α,12β,20(S)-tetrahydroxy-dammar-24-ene (9), pseudoginsenoside Rt3 (10), notoginsenoside R1 (11), ginsenoside Re (12), notoginsenoside N (13), ginsenoside F1 (14), ginsenoside U (15), ginsenoside Rk3 (16), 3β,12β-dihydroxydammar-(E)-20(22),24-diene-6-O-β-D-xylopyranosyl-(1 → 2)-β-D-glucopyranoside (17), ginsenoside Rh4 (18), pseudoginsenoside Rt5 (19), and vinaginsenoside R22 (20) were obtained. Conclusion: Compounds 2, 19, and 20 are isolated from this species for the first time. The 1H-NMR data of compound 19 and 1H-NMR and l3C-NMR data of compound 20 are first reported. Meanwhile, the NMR data of β-D-xylopyranosyl group in compound 9 is corrected. © 2014 Tianjin Press of Chinese Herbal Medicines.

ABSTRACT

Objective: To study the constituents in the dried aerial parts of Eclipta prostrata. Methods: The constituents were isolated and purified by column chromatography and their structures were elucidated by spectroscopic methods (1D, 2D NMR, UV, IR, and HRESI-TOF-MS) and chemical analyses. Results: Eight compounds were isolated and identified as 7-O-methylorobol-4'-O-β-D-glucopyranoside (1), 3'-hydroxybiochanin A (2), echinocystic acid 28-O-β-D-glucopyranoside (3), ecliptasaponin A (4), eclalbasaponin I (5), eclalbasaponin IV (6), echinocystic acid (7), and 3-oxo-16α-hydroxy-olean-12-en-28-oic acid (8). Conclusion: Compound 1 is a new compound and compound 3 is obtained from this genus for the first time. © 2013 Tianjin Press of Chinese Herbal Medicines.

ABSTRACT

To study the chemical constituents of Dipsacus asper, chromatographic methods such as D101 macroporous resin, silica gel, octadecylsilyl (ODS) column chromatographic techniques and preparative HPLC were used, and five compounds were isolated from 70% (v/v) ethanol extract of the plant. By using spectroscopic techniques including 1H NMR, 13C NMR, 1H-1H COSY, HSQC, HMBC and TOF-MS, the compounds were identified as 3beta-hydroxy-24-nor-urs-4 (23), 12-dien-28-oic acid (1), ursolic acid (2), oleanolic acid (3), 3-O-alpha-L-rhamnosyl(1 --> 3)-beta-D-glucopyranosyl (1 --> 3)-alpha-L-rhamnosyl (1 --> 2)-alpha-L-arabinopyranosyl hederagenin 28-O-beta-D-glucopyranosyl (1 --> 6)-beta-D-glucopyranosyl ester (4), 3-O-[beta-D-xylopyranosyl (1 --> 4)-beta-D-glucopyranosyl (1 --> 4)] [alpha-L-rhamnosyl(1 --> 3)]-beta-D-glucopyranosyl (1 --> 3)-alpha-L-rhamnosyl(1 --> 2)-alpha-L-arabinopyranosyl hederagenin (5), separately. Among them, 1 is a new compound, and 2 is isolated from this plant for the first time.

ABSTRACT

A new phenolic amide glycoside, cimicifugamide A (1) along with four known compounds, trans-feruloyl tyramine 4-O-beta-D-glucopyranoside (2), (+)-isolariciresinol 3-O-beta-D-glucopyranoside (3), cimidahurine (4), and 24-epi-7, 8-didehydrocimigenol-3-O-beta-D-xylopyranoside (5) were isolated from the rhizomes of Cimicifuga dahurica. Compound 3 was identified as a lignan and has been obtained from Cimicifuga genus for the first time. The structure of compound 1 was elucidated by IR, UV, HR-MS and NMR spectroscopic methods.

ABSTRACT

Common and variation peak rates were calculated as dual-index for drug samples based on the infrared spectrogram of Angelicae Pubescentis Radix samples from different places. The results indicated that the common peak rates of 4 samples from Sichuan province, 2 samples from Gansu province, and 2 samples from Hubei province were higher than 73% , 79. 5% and 76. 3%, respectively. Also, the variation peak rates of 2 samples from Gansu province were lower than 12. 9%. Therefore, there is similar germplasm resources and similar chemical composition in the samples originated from the same place. However, for the samples originated from different places, their germplasm resources and chemical composition maybe have been changed. Fourier transform infrared spectrometry could be used to evaluate the rule of the fingerprint and overview of Angelicae Pubescentis Radix originated from different places by its changes of germplasm resources and chemical composition.

ABSTRACT

<p><b>OBJECTIVE</b>Conflicting data have been generated from previous studies to determine which kind of relationship exists between HIV-1 specific CD8 Tcell responses and HIV-1 viral load or CD4 count over the course of infection. In this study, 153 HIV-1 infected LTNPs were enrolled to investigate the role of HIV-1 specific CD8 T-cell responses in chronic HIV-1 infection among HIV-1 infected former blood donors.</p><p><b>METHODS</b>The patients were stratified into three groups according to CD4 count: CD4≥500 cells/μL; 350 cells/μL≤CD4<500 cells/μL; CD4<350 cells/μL. PBMCs were isolated from the patients' anticoagulated blood samples. IL-2 and IFN-γ secretions of CD 8 T cells against 17 HIV-1 consensus B full peptide pools were analyzed by using ICS assay.</p><p><b>RESULTS</b>An overall inverse correlation were observed between CD4 count and plasma viral load. Although no significant difference was observed during the comparisons of frequency/breadth of HIV-1 specific CD8 T cell responses, CD4 count stratification analysis showed that different correlation pattern existed in three strata: as for patients whose CD4 counts were less than 350 cells/μL, no significant correlations were identified between frequency/breadth of HIV-1 specific CD8 T cell responses and CD4 count/viral load; as for patients whose CD4 counts ranged from 350 cells/μL to 500 cells/μL, significant correlation was only observed between the response breadth of IL-2+IFN-γ+ CD8 T cells and CD4 count; however, as for patients whose CD4 counts were more than 500 cells/μL, direct correlations were identified between IL-2+IFN-γ+/IL-2+/IFN-γ+ CD8 T cells and viral load or CD4 count.</p><p><b>CONCLUSIONS</b>Universal consistent inverse correlation was only indentified between CD4 count and viral load. The relationship between HIV-1 specific CD8 T cell responses and CD4 count/viral load varied in different CD4 strata, which showed that better preserved CD4 T cells were correlated with better CD8 T cell functions.</p>

ABSTRACT

Several research groups have recently reported that persistent GB virus C (GBV-C) co-infected with human immunodeficiency virus (HIV) leads to slower AIDSs disease progression than HIV-1 infection alone. However, these findings were not confirmed by several other studies. To investigate the association between GBV-C replication and plasma HIV loads and CD4+ T cell counts, 203 HIV-1 positive former blood/plasma donors(FBDs) were enrolled from Fuyang city of Anhui Province in China. Plasma specimens were collected from them and were tested for GBV-C using RT-PCR and ELISA. Out of 203 specimens, 52 (25.6%) cases were positive for GBV-C, including 35 male (67.3%) and 17 female (32.7%) cases. No significant association was identified between GBV-C infection and CD4+ T-cell counts or between GBV-C infection and HIV viral loads. Since all the subjects studied were naive to ART, the influence of therapy on AIDS disease progression was ruled out in this study. Overall, our data indicated that HIV-1 positive male FBDs were prone to be infected, GBV-C coinfection with HIV-1 does not significantly influence HIV/AIDS disease progression during the late stage of chronic HIV-1 infection.

ABSTRACT

<p><b>BACKGROUND</b>Unregulated commercial blood/plasma collection among farmers occurred between 1992 and 1995 in central China and caused the second major epidemic of human immunodeficiency virus type 1 (HIV-1) infection in China. It is important to characterize HIV-1-infected former blood donors and to study characteristics associated with disease progression for future clinical intervention and vaccine development.</p><p><b>METHODS</b>A cross-sectional study was performed on HIV-1-infected former blood donors (FBDs) and age-matched HIV-seronegative local residents. Demographic, epidemiologic, clinical and key laboratory data were collected from all study participants. Both unadjusted and adjusted multivariate linear regressions were employed to analyze the association of the decrease of CD4(+) T-cell counts with other characteristics.</p><p><b>RESULTS</b>Two hundred and ninety-four HIV-1-infected FBDs and 59 age-matched HIV-seronegative local residents were enrolled in this study. The unregulated blood/plasma collection occurred more than a decade (10.8 - 12.8 years) ago, which caused the rapid spread of HIV-1 infection and the high prevalence of co-infection with hepatitis C virus (HCV, 89.5%); hepatitis B virus (HBV) co-infection was observed in only 11 HIV(+)participants (3.7%). Deterioration in both clinical manifestation and laboratory parameters and increase of viral loads were observed in parallel with the decrease of CD4(+) T-cell counts. The decrease of total lymphocyte counts (P < 0.001) and hemoglobin levels (P < 0.001) and the appearance of dermatosis (P = 0.03) were observed in parallel with the decrease of CD4(+) T-cell counts whereas viral loads (P < 0.001) and CD8(+) T-cell counts (P = 0.01) were inversely associated with CD4(+) T-cell counts.</p><p><b>CONCLUSIONS</b>Co-infection with HCV but not HBV is highly prevalent among HIV-1-infected FBDs. CD4(+) T-cell counts is a reliable indicator for disease progression among FBDs. Total lymphocyte counts, hemoglobin level and appearance of dermatosis were positively associated with CD8(+) T-cell counts and viral loads were inversely associated with the decreased CD4(+) T-cell counts.</p>
